Question: Must the AroCell TK 210 ELISA kit be modified for each type of cancer?

Answer: AroCell TK 210 ELISA is a biomarker of cellular proliferation, a common characteristic of cancers, therefore it should be widely applicable.

However, the types of cancer where the AroCell TK 210 ELISA biomarker is most valuable as a diagnostic aid have not yet been established. Clinical trials are ongoing.

Answer: We believe the AroCell TK 210 ELISA kit provides unique new information on patient disease process using a method which is applicable to the technology and instrumentation already in routine use in health care. AroCell TK 210 ELISA results can be combined with those of existing biomarkers of tumor mass (for example PSA and Ca 15-3) to enable more accurate prognoses and better therapy monitoring. Of course, this must be verified in the final results of currently ongoing clinical trials.

Answer: Most tumor biomarkers provide an indication of tumor mass. The potential value of the AroCell TK 210 ELISA serum biomarker provides an indication of tumor growth. Both may be valuable individually and complementary to each other in biomarker panels.

For example, in a study on subjects with breast cancer, the AroCell TK 210 ELISA biomarker showed better discrimination among healthy individuals and subjects with Stage 2 breast cancer than the traditional biomarker Ca 15-3. Importantly, the combination of Ca 15-3 and TK 210 ELISA further increased diagnostic accuracy, demonstrating the value of combining biomarkers that monitor different aspects of tumor growth (1).

Question: What is the best method to store serum samples for AroCell TK 210 ELISA analysis?

Answer: It is recommended to aliquot the samples and freeze them at -20 °C for long-term storage. If the assay is planned to be performed within the 24 h of sample collection, samples can be stored at 4 °C. Avoid repeated freeze-thaw cycles. Prior to assay, frozen samples should be completely thawed and mixed well.

Question: How many samples can be analyzed with the AroCell TK 210 ELISA kit?

Answer:  We recommend running the calibrators, controls and serum samples in duplicates. You can run a total of 40 serum samples.

Question: Which is the best method to plot a standard curve for AroCell TK 210 ELISA analysis?

Answer: We recommend the use of 4-parameter logistic fit (4 PL) for quantitative analysis of the serum samples. Calculate the mean OD for each calibrator, controls, and serum samples from the duplicates. Next, calculate the mean OD for the Calibrator 0 and subtract this value from mean OD values of calibrators (A to E), controls and serum samples. The concentration of the TK1 protein levels can be interpolated from the standard curve.
